CD185 is a G protein-coupled receptor belonging to the chemokine receptor subfamily. Upon binding of its ligand, the chemokine CXCL13, CXCR5 initiates multiple intracellular signaling pathways that regulate cell proliferation, survival, and migration. CXCR5 is expressed in both mature B cells and follicular helper T cells, and respond to CXCL13 gradient to control lymphocyte migration towards secondary lymphoid tissues. CXCR5 has also been shown to be highly expressed in primary breast tumors, in correlation with their propensity to grow and metastasize. CXCR5 binds B-lymphocyte chemoattractant (BLC) and is involved in B-cell homing to follicles in lymph nodes and spleen. Two isoforms are produced by alternative splicing. CXCR5 expression has been reported in human blood and salivary gland and in animal lymphatic organs, blood, bone marrow, brain, liver, lymph node, skin, spleen, stomach, and tonsil. ESTs have been isolated from human brain, germ cell, lung, lymph, pancreas, skin, stomach, and uterus cancer libraries, and from normal human blood, liver/spleen, lymph node, nerve, placenta, small intestine, tonsil, and B-cell/lung/testis libraries.
